controls, primary Abs were omitted and samples were postfixed only in GA, with no UA
before CPD.
Care was taken to record tilt-series of overhanging microvilli (without carbon-formvar support)
and ET procedures completed as described earlier (1).
RESULTS AND DISCUSSION
IET with MEM (1) of freely dangling microvilli gave excellent resolution, and especially after
postfixation with GAOsTA (Figs. 1, 2) revealed a structurally highly detailed appearance with
detectable 1.4 nm gold particles labelling ezrin (Figs. 2, 2a). The resolution was superior in
comparison to preparations with 3D-reconstructions achieved by weighted-back-projection
(WBP) method (Fig. 3) or without TA (Fig. 4). We could also detect that 5 nm and 10 nm
gold particles labelling mucin or ezrin (Figs 2, 4), respectively, appeared grainy indicating
substructural details (Fig. 2b). We are planning to correlate by superimposing the 3D-
reconstructions of isolated ezrin with the IET data from microvilli.
